    Retroviruses such as HIV-1 replicate in the nucleus, but the nuclear entry mechanisms of other retroviruses remain poorly understood. This project focuses on Mason-Pfizer monkey virus (MPMV), which represents a promising and safer model for gene therapy, to investigate whether intact capsids transit through the nuclear pore and if subsequent nuclear actin-myosin transport is required for integration. Using super-resolution fluorescence and electron microscopy we will visualise GFP-tagged MPMV capsids in infected cells. We will use targeted mutagenesis of the capsid gene and known HIV-1 capsid inhibitors to determine whether MPMV shares pathways with HIV-1. Reverse transcriptase-deficient mutants will determine whether integration depends on reverse transcription before nuclear entry. By combining co-immunoprecipitation, mass spectrometry and cytoskeletal inhibition, we will map key virus-host nuclear interactions. These findings could reveal common retroviral import strategies, and provide insight into the design and development of more efficient retroviral vectors for gene therapy.
