    Charakterizace Fur fenotypu mutantů Pseudomonas aeruginosa: nadprodukce komponent transportního systému pro FeIII.

    Projekt se zabývá charakterizací transportního mechanismu pro FeIII, jednoho z faktorů virulence, u Fur mutantů P. aeruginosa a možným využitím těchto poznatků v biotechnologiích. Projekt řeší následující cíle: 1. vztah mezi nadprodukcí pyoverdinu a jehospecifického receptoru u Fur mutantů, 2. maximální biosyntetickou kapacitu pro komponenty FeIII-transportního systému v rekombinantních kmenech a Fur mutantech a 3. charakterizaci změn ve Fur regulonu u rodičovského a Fur fenotypu. Metodami rekombinantní DNK budou připraveny rekombinantní kmeny obou fenotypů, u nichž bude sledována nadprodukce komponent FeIII-transportního systému za definovaných kultivačních podmínek v míchaném bioreaktoru. Pomocí metody DNA microarray budou studovány změny ve Fur regulonu u rodičovského a Fur fenotypu jako odpověď buněk na přítomnost vysoké koncentrace FeIII v mediu. Nadprodukované komponenty transportního systému lze využít v medicíně, farmacii a dalších oblastech vědy a výzkumu.
