Mechanisms of Neuropathic Pain States
Project goals
The aim of this project proposal is to study the influence of synaptic transmission modulation of nociceptive signalling at the spinal cord level in neuropathic pain states. Neuropathic conditions will be studied using models of peripheral and spinal nerve injury and also after application of platinum chemotherapeutics. The introduction and investigation of chemotherapy induced model of neuropathy will be done in cooperation with professor Dougherty (USA) and will be also one of the major goals of thiscooperation. The role of presynaptic TRPV1 receptors modulation on the endings of primary afferents in the spinal cord will be the main focus of this study. Learning the mechanisms of nociceptive modulation in this project may help to determine new targets for therapy of neuropathic pain patients.

Cílem tohoto projektu je zjistit vliv modulace synaptického přenosu na míšní úrovni na rozvoj stavů neuropatické bolesti. Budou studovány neuropatické stavy na modelech po poškození míšního a periferního nervu a také po aplikaci platinových chemoterapeutik. Zavedení a studium modelu neuropatie po aplikaci chemoterapeutik ve spolupráci s profesorem Doughertym v USA bude také jedním z hlavních cílů této dvoustranné spolupráce. Na modelech neuropatie bude studována především úloha modulace presynaptickýchTRPV1 receptorů na primárních aferentních zakončeních v míše. Objasnění mechanizmů modulace nociceptivního přenosu v tomto projektu, může přispět k získání nových přístupů léčby pacientů s bolestmi neuropatického původu.

In the fourth year of the project LH12058 (Mechanisms of neuropathic states development) with principal investigator Jiri Palecek MD, PhD at the Institute of Physiology Academy of Sciences of the Czech republic vvi, the importance of spinal TRPV1 and TLR4 receptors in the modulation of neuropathic pain states induced by chemotherapeutic paclitaxel was confirmed.
