    The objectives of the project are divided into three areas according to their scientific nature. 1) Synthesis. The objectives in this area are 1a) The synthesis of a complete library of systematically deoxofluorinated deprotected analogs of LacNAc, LacdiNAc and diacetylchitobiose ready for determination of binding affinity, and for NMR epitope mapping; 1b) The synthesis of systematically deoxofluorinated GlcNAc, GalNAc, and ManNAc protected at hydroxyl groups as acetyl, propionyl and butyryl esters; 1c) Preparation of a limited number of protected disaccharide units of GAGs intended as a demonstration of the feasibility of this first step towards assembly of functional segments of fluorinated GAG. 2) Ligand recognition. The objectives in this area are 2a) Determination of the binding affinity of deoxofluorinated LacNAc, LacdiNAc and diacetylchitobiose analogs towards their cognate lectins by ELISA and ELLA assays; 2b) Epitope mapping of LacNAc, LacdiNAc and diacetylchitobiose using advanced NMR techniques. 3) Metabolic inhibition. The objectives in this area are 3a) Identification of deoxofluorinated acylated hexosamines usable for manipulation of cell-surface glycome by metabolic inhibition; 3b) Determination of how the cell-surface glycome has been altered after treatment with compounds identified under 3a.

    The project included synthesis and lectin affinities of three complete series of fluorinated disaccharides, synthesis and cytotoxicity of fluorinated amino saccharides, their organometallic conjugates, NMR analysis of interactions with galectin-1 and -3, and conformational analysis. Three publications in WoS-indexed journals were published and two more are being prepared for submission.
