    Nuclear regulatory landscape of bZIP transcription factors in plant reproductive development (REPROZIP)

    The principal objective of this project proposal is to decipher and visualise the complexity of pollen Group E/I bZIP TF nuclear regulatory landscape in relation to chromatin dynamics during Arabidopsis pollen development and under the effect of environmental cues. For this, we will take the advantage of our model material being effectively single cell system – naturally synchronised population of developing spores and growing pollen tubes. To demonstrate the function of bZIP regulatory landscape in pollen it will be necessary to visualise bZIP proteins and their interactions and to identify their downstream pathways, target genes and binding sites. This will be achieved by the functional characterization of bzip double mutants and by the combination of transcriptomic (RNA-seq), proteomic (LC-MS/MS) and lipidomic (LC-MS) approaches that will be compared to the previously published bzip34 and bzip18 microarray data. As all bZIP genes of interest were demonstrated to interact in the Y2H assay, we aim to clarify whether they co-operate on the same regulatory pathway(s) and whether they form native functional and/or regulatory complexes.
