    Two for the Price of One: Illuminating the Dual Role of Paralogous Exocyst Subunits in Plants

    Exocytosis, the final step of the secretory pathway, is spatiotemporally controlled by the exocyst, an evolutionarily conserved protein complex. Uniquely in plants, several exocyst subunits diversified into multiple paralogs. We recently discovered that sporophyte- and pollen-dominant isoforms of these subunits can exhibit non-redundant dual functions, making the exocyst an exciting model that bridges cell biology and evolution. We hypothesise that this adaptive diversification of exocyst subunits introduces new layers of regulatory control over vesicle trafficking. Focusing on paralogous EXO84 and SEC15 isoforms that display dual roles, we aim to define how their structural and biochemical properties confer specificity in both sporophyte and pollen. By employing multifaceted methods, we will dissect exocyst architecture in different tissues, clarify isoform relationships, and identify novel interactors that govern exocyst function. This research will enrich our understanding of membrane traffic evolution in plants and beyond.
