Biomolecules are the fundamental building blocks of every organism. Yet, there is still a myriad of molecular processes that remain elusive, creating significant roadblocks in, for example, medical research. Biomolecules possess heterogeneities of high physiological relevance that can only be unravelled using single-molecule tools. However, the available tools often do not provide quantitative information and require fluorescent labelling or immobilization onto a surface, possibly altering the biomolecule’s natural behaviour. Dioscuri Centre (DC) will focus on developing next-generation life-science tools that can bypass those limitations and thus significantly contribute to the fundamental understanding of molecular processes pervading biological systems. DC will develop a unique experimental toolbox for multiparametric characterization of biomolecular processes at a single-molecule level without the need for any labelling or attachment onto a surface. The tools will build on the principles of Nanofluidic Scattering Microscopy - a ground-breaking optical microscopy technique that has been recently introduced. The developed tools will be used for various applications, including investigation of biomolecular interactions, aggregation processes, molecular transport, or single-cell analysis. The research will inspire new strategies for early diagnosis and therapeutic intervention, plus the invention of new biotechnologies mimicking nature.
