Perfect markers for resistance genes in pea (Pisum sativum L.) to Fusarium root rot and Fusarium wilt .
Project goals
Direct markers are the next evolution in crop improvement strategies. Modern plant breeding programs employ DNA markers linked to the trait of interest and PCR technologies to quickly identify lines to improve efficiencies in releasing agronomically superior cultivars for pea crop production. With the advent of cloning the genes/alleles controlling economic traits like quality and disease resistances, traits can be ed directly using PCR technology. We proposed to fine map the loci we have identified forresistance to Fusarium root rot and Fusarium wilt, using bulk segregant analysis of recombinants flanking important resistance loci for each disease. For fine mapping, candidate genes will be utilized from sophisticated plant-microbe molecular interaction studies. Validation of the candidate gene(s) effect (s) will be by association mapping in a germplasm population previously phenotyped for reaction to Fusarium root rot and Fusarium wilt.

All three QTL loci (FRR1-2-3) responsible for Fusuarium solani resistance were mapped using microsatellite and gene specific markers. pI39 was confirmed as candidate gene for FRR2 locus, while DRR206 was rejected. Fw locus responsible for Fusarium oxysporum rasa 1 resistance was also mapped with higher precision on LGIII. Pea accessions were phenotyped for both fusarium pathogens.
