Annotation in Czech
A number of studies, including our own preliminary data, have shown that TRAIL (an important member of the TNF family) works synergistically with some anticancer drugs to kill most types of tumor cells and reversed resistance to such drugs. These findings indicate that the combination of TRAIL and chemotherapeutic drugs is potentially promising in treating refractory cancers. TRAIL can induce human cancer cell apoptosis through engagement of its death receptors DR4 and DR5. However, not all cancers aresensitive to this cytokine. Recently, it was shown that low doses of cytotoxic drugs could restore TRAIL-induced cell death in resistant colon and prostate cancer cell lines. Taken together, finding mechanisms leading to sensitizing the cancer cells to chemotherapy represent a useful approach for identifying new and/or improving existing anti-cancer strategies. Project results evaluation
The main contribution was on the methodological level. The results demonstrated that Pt-cytostatics may modulate signalling of TRAIL by different mechanisms - changes of cell membrane, expression of receptors and kinetics of internalization of TRAIL.
