The detection of biofilm as a factor of virulence within complex diagnostics of yeast blood stream infections

Project goals

The aim of the project is to put the theoretical knowledge about yeast biofilm into clinical praxis and to evaluate the methods of rapid diagnostics of yeasts (PCR, detection of the specific antigen). Strains of the genus Candida isolated from human material (neutropenic patients and patients with foreign-body infections) will be studied. The values of minimal inhibitory concentration (MIC) and minimal biofilm inhibitory concentration (MBIC) of different antomycotics will be determined and statisticallycompared. The applicability of MBIC assessment into the routine laboratory praxis will be evaluated.

    Projekt je zaměřen na zavedení teoretických poznatků o biofilmech do klinické praxe a na posouzení významu metod rychlé diagnostiky. Studie zahrnuje kmeny kvasinek izolovaných z humánního materiálu (pacienti s neutropenií a umělými implantáty). Budou ověřeny možnosti rychlé detekce kvasinkových infekcí (PCR, průkaz antigenu) a bude stanovena minimální inhibiční koncentrace (MIC) a minimální biofilm inhibující koncentrace (MBIC) a bude provedeno jejich statistické srovnání a zhodnocení. Dále bude ověřenapoužitelnost navrhovaného postupu ke stanovení MBIC a studiu biofilmů v laboratorní praxi.

    The detection of candidaemia usig PCR or mannan has limited value. The ability to produce the biofilm correlates in C.parapsilosis well with clinical significance. The biofilm forms are more resistant;resazurin is a useful tool in sensitivity testing.
