Sleep and brain changes related to memory decline in mild cognitive impairment

Project goals

The expected aging of the population will increase people with memory disease and dementia, especially Alzheimer's disease. Mild cognitive impairment (MCI) is a risk syndrome in which intervention can delay the development of Alzheimer's disease. Sleep disorders and changes in magnetic resonance imaging of the brain occur along with memory impairment in MCI. The aim of the project is to find the connection between slow-wave sleep and sleep spindles, corresponding brain structures and memory consolidation during sleep using polysomnography, magnetic resonance imaging of the brain and paired associative learning test. Then we will investigate whether it is possible to enhance memory consolidation during sleep in MCI by manipulating sleep rhythms using acoustic stimulation. Based on the results, we will illuminate mechanisms between sleep, brain structure, and memory impairment in MCI. This comprehensive sleep approach can bring new insights into early interventions and other clinical strategies at the stage of MCI.

    Změny spánku a mozku ve vztahu ke zhoršení paměti u mírné kognitivní poruchy

    S očekávaným stárnutím populace bude přibývat osob s poruchou paměti a demencí, především Alzheimerovou nemocí. Mírná kognitivní porucha (MKP) je rizikovým syndromem, v němž intervence může oddálit rozvoj Alzheimerovy nemoci. Souběžně se zhoršováním paměti u MKP se objevují poruchy spánku a změny na magnetické rezonanci mozku. Cílem projektu je zjištění souvislostí mezi pomalovlnným spánkem a spánkovými vřeténky, odpovídajícími mozkovými strukturami a konsolidací paměti během spánku pomocí polysomnografie, magnetické rezonance mozku a párového asociačního testu učení. Poté budeme zjišťovat, zda je možné konsolidaci paměti během spánku u MKP zlepšit ovlivňováním spánkových rytmů pomocí akustické stimulace. Na základě výsledků osvětlíme mechanismy mezi spánkem, strukturou mozku a poruchami paměti u MKP. Tento komplexní přístup zaměřený na spánek může přinést nové poznatky pro časné intervence a další klinické strategie ve stadiu MKP.

    The main goals of the project were to monitor the connections between sleep, brain structure and memory disorders, specifically: 1. to investigate the effect of sleep on memory impairment in amnestic mild cognitive impairment and to demonstrate that the memory consolidation can be improved by acoustic stimulation during sleep 2. to determine whether it is possible by specific targeted manipulation of sleep rhythms in patients with amnestic mild cognitive impairment to improve memory consolidation. The project was disrupted by a number of objective reasons; however, the authors were able to complete data collection from the required number of volunteers despite these limitations. Partial results of the project (improved methodologies, tests, etc.) were published in a number of international journals with IF (their number is several times higher than the original amount stated in the application).
