    Hmotnostně spektrometrické analýzy rekombinantních proteinů

    Předmětem řešení projektu bude hmotnostně spektrometrická analýza (provedená na pracovišti dodavatele) vzorků připravených žadatelem. Cílem projektu bude identifikace a charakterizace vzorků. V rámci tohoto projektu budou prováděny tři typy analýz: 1) Identifikace rekombinantního proteinu a určení jeho primární struktury 2) Identifikace rekombinantního proteinu s detekcí fosforylace a určení místa fosforylace 3) Identifikace rekombinantního proteinu s detekcí glykosylace a následnou charakterizací glykanové části.

    Two recomb. proteins produced within the GENERI BIOTECH s.r.o. (GB), hSHBG and sGNGB1 protein. Proteins in different fractions were passed to FVZ UO for SDS-PAGE analysis. In the FVZ UO, protein separation was performed on SDS-PAGE and the isolated proteins were subjected to further analyzes. Protein identification was performed based on the composition of peptide fragments. Positive hSHBG and sHMGB1 were identified. An analysis of post-translational modifications was carried out, where sugar residues were identified. The co-operation procedure between GB and FVZ UO has been optimized for the analysis of recomb. proteins to be used. The results of the hSHBG and sHMGB1 protein analyzes were important for further development of protein production for their commer. use. It is the confirm. of the identity of the proteins and their purity and the confirm. of the presence of glycosylation, which is important for application of proteins produced in mammalian systems.
