    We tested a variety of physical and chemical non-vector based transfection methods in order to evaluate which of them gave the most suitable transfer of siRNA into chronic myeloid leukemia (CML) primary cells. Chemically synthesized siRNAs against several genes were transfected into the cells and level of knockdown was determined by qRT-PCR. According to our results, nucleofection (Amaxa) appears to be the only suitable non-viral method for siRNA delivery into the hard-to-transfect CML primary cells

    Za účelem vybrání nejvhodnějšího postupu vnášení siRNA molekul do primárních buněk chronické myeloidní leukemie (CML) byla testována škála fyzikálních a chemických non-vektorových transfekčích metod. Chemicky připravené siRNA proti několika genům byly transfekovány do buněk a účinnost transfekce byla určena pomocí qRT-PCR. Na základě našich výsledků bylo zjištěno, že nukleofekce (Amaxa) je jedinou účinnou metodou pro transfer siRNA do těžko transfekovatelných CML buněk
