Ring test results of STS markers for leaf rust resistance genes in wheat

  • Original language description

DNA markers: STS, SCAR, CAPS and SSR, for 18 1eaf rust resistance genes in wheat are updated and primers for their amplification are listed. Five selected STS markers for leaf rust resistance genes Lr9, LrlO, Lr19, Lr24 and Lr28 were identified in the network of seven Euro- pean laboratories in 7 accessions, having the above mentioned genes. Near-isogenic lines of spring wheat Thatcher were used as positive controls. Markers for resistance genes Lr9, LrlO, Lr19, Lr24 were identified in all the seven laboratories as amplification products 1100 bp, 310 bp, 130 bp and 310 bp, respectively. The STS markers linked to resistance genes Lr9, LrlO, Lr19, Lr24 were found to be highly specific for these genes and will be useful in marker-assisted selection in wheat. However, amplification product 378 bp for resistance gene Lr28 was detected in all the accessions in all the seven laboratories, both in the accessions having the gene and those not having it. This marker needs to be improved.
