    Molecular basis of acetolactate synthase inhibitor resistance in Czech biotypes of kochia

    Seven biotypes of kochia (Kochia scoparia (L.) Schrad.) resistant to chlorsulforon have been found in the Czech Republic since 1996. Several characteristics of these biotypes have been investigated so far. This is the first study on the molecular basis of their herbicide resistance to acetolactate synthase-inhibiting herbicides. Eight biotypes of kochia were examined for correspondence between restricion fragments of PCR amplification products and phenotypic resistance to chlorsurforon of individual plants. Two regions of the gene encoding acetolactate synthase (ALS) were amplified, digested with restriction enzymes and sequenced. The resistance to ALS-inhibiting herbicides in all Czech kochia biotypes studied was conferred by a mutation at codon 574 of the ALS gene. Analysis of the results of DNA sequencing indicated that the mutation induced a leucine for tryptophan substitution.
