    Comparison of Detection Methods for Acarus siro (Acari:Acaridida: Acarididae) Contamination in Grain

    Acarus siro L. (Acari: Acaridida: Acarididae) is an important pest of stored grain because it contaminates the grain by allergens and transfers pathogenous microorganisms. Rapid detection of contamination enables to intercept an early grain infestation by the pest. In this study, we compared the usability and efciency of various detection approaches. Under laboratory conditions, grain samples of various sizes were infested by different levels of the following contaminants: eggs, adults, and feces of A.siro. The samples were analyzed by enzyme-linked immunosorbent essay (ELISA) by using anti-A. siro polyclonal antibody (immunochemical method), extracted in Berlese-Tullgren funnels, sieved, and processed by the flotation (conventional methods). The adults or juveniles of A. siro could be detected by all the three tested conventional methods and ELISA with detection limits in the range from 221 to 1,157 mites/kg grain.     Práce se zabývá porovnáním detekčních metod pro identifikaci kontaminace roztočem Acarus siro. Jsou testovány klasické detekční metody (extrakce na Berlese-Tullgren extraktoru, prosévání a flotace). Další použitou metodou byla ELISA s nově připravenou polyklonální protilátkou pro detekci A. siro. Bylo zjištěno, že dospělce a juvenilní stádia bylo možné detekovat všemi uvedenými metodami s rozdílným detekčním limitem v rozsahu 221 až 1157 roztočů/kg obilí. Vajíčka bylo možné detekovat pouze pomocí flotace (detekční limit 1950 vajíček/kg obilí). Exkrementy roztoče byly detekovatelné pouze pomocí imunochemického testu s detekčním limitem 1.04 ?g exkrementů/g obilí, což znamená, že použitá technika umožní detekci jednoho metabolicky aktivního roztoče/g obilí.
