Original language description
Applicability and conditions of in vitro cultures for testing of pome fruit (apple, pear) resistance against fire blight were tested. MS medium according Murashige and Skoog (1962) with addition of different cytokinin concentrations proved to be suitablefor in vitro cultivation. We obtained multiplication coefficient higher than 1.5 for all apple and pear genotypes. Isolate of bacterium Erwinia amylovora (Ea 8/95) was verified. Three methods of artificial inoculation of in vitro cultures were tested. Decapitation of apical part of the shoot tip by scissors and applying of inoculum (3 ?l) by pipette on the cut area (variant 1) proved to be the best method for artificial inoculation. Preliminary results of inoculation of in vitro cultures by Erwinia amylovora indicate the possibility of using of the method for testing of pome fruit resistance against fire blight.
