Background: Sodium channels (SC) in mites and insects are target sites for pesticides including pyrethroids. Point mutations of SC gene were reported to change structural conformation of the protein and sensitivity to pesticides. To find mutations in SCgene of Varroa destructor (VmNa) we analyzed VmNa gene sequences available in GenBank and prepared specific primers for amplification of two fragments harboring the regions coding for (i) IIS4-IIS6 in positions 2837-3337 and (ii) IIIS4-3´terminus in positions 4737-6500 according to VmNa cDNA sequence AY259834. Results: Sensitive and resistant populations did not differ in amino acid sequences of the IIIS4-3´terminus VmNa region. The differences were found in the fragment IIS4-IIS6: In resistant population, mutation C(3004)>G resulted in substitution L(1002)> (codon ctg > gtg) in the position equivalent to L925 in house fly in the linker connecting segments IIS4 and IIS5.
