Detection and quantification of Puccinia graminis and Piccinia tricina in host plant tissue by real-time PCR
Original language description
Leaf rust, caused by Puccinia triticina Eriks., is wheat's most common rust disease (Triticum aestivum L.) in more worldwide regions. Puccinia graminis Pers. is one of the other most devastating diseases of cereal crops grown worldwide, often resulting in significant yield losses. This work aims to determine the quantity of the given rust in training populations of Triticum aestivum cultivars bred for resistance to rusts.Tubulins are well conserved in eukaryotes and are molecular targets of MBC fungicides, effectively controlling many plant diseases caused by ascomycete fungi. Therefore, the B-tubulin gene was selected as a molecular marker, and primers and probes were developed for it. The symptomatic evaluation was performed using image analysis and RT-PCR quantified pathogen content.By applying genomic selection, the material with the best combination of the observed traits associated with resistance to these rusts in the population will be selected.
