    Prospects of using surfactants in cryopreservation of plant genetic resources

    Effective saturation with cryoprotectants is extremely important for the preservation of plant germplasm during cryoconservation, especially when using highly concentrated PVS. Reducing the surface tension of solutions by adding surfactants is actively used in agriculture to wet hydrophobic and hard-to-reach plant surfaces covered with a waxy layer and a dense coating and to accelerate the absorption and distribution of the solution in plant tissues. However, there are no data on the use of surfactants to accelerate the saturation of plant genetic resources with cryoprotective solutions. The purpose of the study was to evaluate the effectiveness of the use of surfactants in cryoprotective media to accelerate the saturation of plant explants. In the study, 100, 80, and 60% solutions of PVS2 with the addition of organosilicone surfactant Siliksan106 in concentrations non-toxic to plants (0.025÷0.15%) were investigated. Using stalagmometry, it was found that the addition of surfactants leads to a 3-fold (depending on concentration) decrease in the surface tension of PVS2 solutions. The osmotic reactions of cells after the addition of PVS2 were studied under a microscope using the Chlorococcum dissectum microalgae as an example, and a significant decrease in the time of cell volume recovery in the presence of surfactants was found. The use of surfactant during the saturation of grape cuttings (Frontinak variety) by the vacuum infiltration method reduced the time of PVS2 passing through the capillary system of the cuttings by more than 3 times. DSC study of low-temperature phase transitions of PVS2 solutions did not reveal significant differences when surfactant was added. Thus, the use of surfactant in the composition of cryoprotective solutions accelerates the cellular and capillary permeability of plant explants for cryoprotectants, without reducing their glass-forming ability. The results prove the great potential of surfactants in the plant cryopreservation.
