Currently, a number of effective plant vitrification solutions (PVSs) have been developed but they contain high concentrations of cryoprotectants with limited incubation time. Changing the incubation temperature and PVS concentration makes it possible to regulate not only cytotoxicity, but also the viscosity of solutions, which affects the dynamics of plant saturation with cryoprotectants. The purpose of this work was to determine the influence of viscosity changes on the dynamics of microalgae' saturation with PVSs. Viscosity in PVSs of different concentrations was investigated in the temperature range from 1oS to 30oS by the capillary viscometer method. It has been shown that the viscosity increases in the series PVS1, PVS2, PVS4, PVS3, which correlates with an increase in the concentration of glycerol, as the most viscous component. Due to the high concentrations of glycerol, the viscosity of 88% PVS3 was 35-48 times higher than that of 100% PVS1, PVS2, PVS4, and when the PVS3 concentration decreased from 88% to 60%, the viscosity decreased by more than 50 times at 1oC. An increase in temperature decreased the viscosity in all investigated solutions. For example, when the temperature changed from 1oC to 30oC, the viscosity of 100%, 80%, 60% PVS2 decreased by 4.5, 3.5 and 3 times, respectively. A correlation was found between the viscosity of PVSs and the dynamics of their penetration into microalgae Chlorococcum dissectum. It was shown by microscopy that after dehydration in 60% PVS2, the cell volume of 90% cells was recovered, and in 80% and 100% PVS2, the number of recovered cells was 33% and 27% after 5 min of incubation. Thus, by varying the viscosity of PVSs, it is possible to effectively influence the dynamics of their permeability to plant cells.
