    Isolation and characterization of chitosan - glukan complex from Aspergillus niger biomass

    Chitosan and chitin -glucan complex were extracted from Aspergillus niger mycelial wastes of citric acid production.Mycelium was treated with concentratedaqueous NaOH solution (40%) at 130°C for 1-3 h. On applying this procedure, the insoluble material is left as a white powder (15 - 33,5 % on a dry cell basis). For chitosan isolation, this freeze-dried alkali insoluble fraction was treated with diluted HCl solution (2 %) at 95°C for 15-20 h. The yields of the acid soluble chitosan fraction were 5-15 %on a dry cell basis on this condition. IR spectrometry analysis confirmed that the acid soluble material contains chitosan and the acid insoluble remainder consists of chitin and glucan. Results of nitrogen analysis showed a significant prevalence of chitin in the acid insoluble remainder. An original fast and simple method of degree of deacetylation determination was developed using direct UV spectroscopy. Degree of deacetylation of chitosan in the acid soluble fraction is at least 95 %
