    Induced polyploidization in Moringa oleifera using oryzalin

    Moringa oleifera Lam. (drumstick tree) is a fast-growing deciduous tree of the family Moringaceae, native to the Indian subcontinent. Moringa trees are grown mainly in semiarid, tropical, and subtropical areas. It has multiple uses due to its rich content of bioactive compounds and nutrients and its adaptability to a wide range of agroclimatic conditions. Moringa is referred to as “a miracle tree”; therefore, it is desirable to obtain new genotypes with original properties. The aim of this study was to obtain tetraploids using induced in vitro polyploidization by treatment with an antimitotic agent. In vitro shoots were shaken in liquid full-strength Murashige and Skoog (MS) induction medium with oryzalin added at a concentration 50 µM and 100 µM for 24 h or 96 h of cultivation. The ploidy level was confirmed by flow cytometry. Then, tetraploids were compared with diploids in terms of basic morphological and physiological characteristics. It was found that the oryzalin concentration and exposure time have no effect on the induction of tetraploid plants. On the other hand, both factors influenced the regeneration of the in vitro culture. Tetraploids were obtained in all treatments, but the efficiency was low. The ploidy level had a significant effect on plant growth and morphology.
