Original language description
PALM microlaser technology was used to generate bovine, porcine and equine chromosome paint probes. Single chromosomes were isolated by laser dissection of the membrane around the target chromosome and 10-15 copies of the chromosome were collected by catapulting them into the Eppendorf cap. The microdissected DNA was amplified by degenerate oligonucleotide-primed PCR. The amplification product was labelled with digoxigenin and spectrum orange in secondary PCR, or by Biotin Chem-Link, and hybridized ontometaphase spreads. Fluorescence in situ hybridization signal revealed specific painting of the chromosome. Paint probes generated by this method are presented.
