    11th International Conference Nanotechnology for Healthcare: Progress in Recombinant Vaccines, Molecular Adjuvants and Modern Drug Delivery Systems, Telč, 5.-7.6.2018 – lecture. Introduction: Purpose of the study was to prepare highly monodisperse polymeric nanoparticles which could be used as a model to study the effect of particles size to oral mucosa penetration capabilities in/within sublingual vaccination formulations/systems. Methods: Herein model poly lactic-co-glycolic acid (PLGA) nanoparticles were prepared by adapting the so-called nanoprecipitation or solvent displacement process to commercially available microfluidic mixing platform NanoAssemblr (Precision Nanosystems, Canada). Resulting nanoparticles were characterized by Dynamic light scattering (DLS), Nanoparticles Tracking Analysis (NTA) and transmission electron microscopy (TEM) Results: By systematic optimization of main process parameters as total flow rate (TFR), flow rate ratio (FRR), solvent hydrophobicity and polarity, polymer concentration, temperature and stabilizer concentration, the uniform particles were produced (PDI ≤ 0.009) with good reproducibility over wide range of sizes (<50 to >200 nm). Also, the effect of individual process parameters on the shift of ternary phase diagram boundaries was studied in order to understand and clarify particles formation mechanism. Discussion: Obtained results demonstrated that control of mixing kinetic provided by NanoAssembler platform is crucial for an increase of nanoparticles formulation quality in terms of their physicochemical characteristic (size and polydispersity). The quality of prepared nanoparticles was more than sufficient to use them for further study within our new mucoadhesive nanofibre-based sublingual vaccination patch system. Hence it can be assumed that microfluidics will become an indispensable tool for further development on the field of nanomedicine in the future.
