    Detection of African swine fever virus by xMap technology

African swine fever virus (ASFV) is causative agent of hemorrhagic fever, an emerging infectious disease with high mortality rate in domestic pigs and wild boars. Humans are not susceptible to the disease. The virus is responsible for serious problem with production and economic losses in many countries. The infection can be spread by direct contact with infected animals, and also via pork products contaminated feed and fomites (e.g. shoes, clothes, vehicles, knives). Early diagnosis of the disease and detection of the virus could help prevent spreading of ASFV between animals and through the countries. Our research is focused on rapid multiplex systems for detection of swine pathogens with special emphasis on ASFV. Methodical part is based on the multiplex oligonucleotide ligation – PCR (MOL-PCR). The first step is multiple oligonucleotide ligation of specific probes, designed for the gene VP72 and central variable region - B602L of ASFV. Highly conserved region of gene VP72 is detectable in all 23 genotypes of ASFV and B602L is used to differentiate between several genotypes. Ligation is followed by amplification, hybridization of PCR products to magnetic microspheres and finally signal detection on xMap (Luminex, Texas). Detection system was optimized on avirulent strain of ASFV BA71V. Subsequently the specific probes were validated on biological samples (spleen) originating from infected wild boars in the Czech Republic. Our concept for detecting of ASFV by MOL-PCR shows potential for diagnostic application in combination with the probes directed to the other porcine pathogens to create a complex diagnostic method.
