    Immunomodulatory potential of Lacticaseibacillus rhamnosus-derived extracellular vesicles

    The key effects of the gut microbiota on host physiology and immunity are, in part, mediated by extracellular vesicles (EVs) released by these bacteria. The gut microbiota-derived EVs were found in human blood and urine, demonstrating how bacterial EVs (bEVs) can be distributed locally or systemically throughout the human body. The aims of the study were biophysical and biochemical characterization of bEVs from Lacticaseibacillus rhamnosus CCM7091, originally isolated from human faeces, and determination of their immunomodulatory potential. Two populations of particles (30-40nm; 90-120nm) with a concentration of 1013 particles per ml were detected. The purified bEVs samples contained 200-300ug/ml proteins per bEVs pellet with similar protein profiles determined by SDS-PAGE. A total of 337 proteins were consistently identified in bEVs prepared under the same cultivation conditions followed by standard isolation and purification protocol, with the most proteins belonging to the environmental information processing group. Overall, the first results showed pro-inflammatory action of Lacticaseibacillus-derived bEVs, stimulating the formation of reactive oxygen and nitrogen species as well as production of pro-inflammatory cytokine TNFalfa by RAW264.7 macrophages. Lacticaseibacillus rhamnosus CCM7091 produces EVs with similar protein profiles under standardized cultivation conditions. The first insight into their immunomodulatory potential showed overall pro-inflammatory action.
