Common carp (Cyprinus carpio) is an economically important species highly susceptible to spring viraemia of carp (SVC) infection caused by Carp sprivivirus (SVCV, Rhabdoviridae). The invasive gibel carp (Carassius gibelio) is considered susceptible to SVCV, but to a lesser degree. Despite the presence of natural hybrids of these cyprinid species in the waters of continental Europe, knowledge about their immune response to SVCV infection are lacking. In this study, common carp (n¼22), gibel carp (n¼17) and their F1 hybrids (n¼15) were intraperitoneally injected with Carp sprivivirus strain CAPM-V539 with a dose of 8,6x106 TCID50 per fish. Blood serum was collected at 20, 35, 50 and 100 dpi and examined for the presence of specific antibodies against SVCV. Custom ELISA consisted of purified SVCV CAPM-V539 antigen, monoclonal mouse against carp antibody 1E10/2A8 and a commercial horseradish peroxidase-conjugated antimouse antibody. Absorbance was measured at 450 nm and expressed as the difference of values of the positive and the negative antigen adjusted for the mean value of absorbance of a standard positive control. Common carp showed the highest specific IG values, culminating at 35 dpi with absorbance of 0.62. Hybrids produced significantly less specific antibodies, with peak absorbance value of 0.31. Lastly, with a mean absorbance never exceeding 0.007, gibel carp did not show presence specific SVCV IG above the levels measured in negative control at any stage of the trial.
