The study aims to characterize Lactic Acid Bacteria (LAB) with potentially probiotic features derived from the gastrointestinal tract of domestic pigs and wild boars. In the future, selected isolates with the best probiotic properties should be used as a component of a probiotic mixture to protect domestic piglets from post-weaning diarrhea. Besides the phenotypic testing of LAB isolates, we also performed genotype characterization using Next Generation Sequencing (NGS) by Illumina. Selected representatives were sequenced to find genes encoding beneficial probiotic properties (adhesion-associated genes or CAZy genes). For sequencing data validation, we use tools like Prokka for annotation, Fast-ANI for strain identification, MEGA-X, MAUVE, and Easy Fig for data visualization. Based on appropriate potential probiotic properties, six isolates were selected. These representatives were cultured in special conditions and mixed into probiotic mixtures. After that, we performed two in vivo experiments on post-weaned piglets in experimental stables in our institute. In the first experiment, we tested the functionality of the three different probiotic mixtures (A, B, C) and the efficiency of the two types of starter feed. During 42 day long experiment, we collected data by observing animal increments, nutrient conversion, and hematological parameters. The second in vivo experiment was conceived as infectious. Every group of testing piglets was given an infectious dose of EPEC (enteropathogenic E. coli) to test the functionality of probiotics after infection. After infection, the changes in the clinical state of piglets were observed, and increments and nutrient conversion were measured. As with the non-infectious experiment, blood and stool from piglets at various intervals after infection were collected (data not evaluated yet).
