    Penile squamous cell carcinoma (pSCC) is a rare genitourinary tumor associated with poorer prognosis in comparison to more common urological cancers. Elst and colleagues report on a novel single-cell RNA sequencing analysis that identified TP53 mutations as a crucial marker of aggressive cancer, independent of human papillomavirus (HPV) status. TP53-mutated pSCCs exhibited epithelial-mesenchymal transition (EMT) transcriptional signatures. Morphologically, this was evident as high tumor budding, which is a well-known indicator of metastasis and poor prognosis across various cancers. Our group recently described the negative prognostic impact of tumor budding, mutated p53 expression profiles (n = 152), and high tumor mutational burden (n = 136) in relatively large cohorts.
