Determination of adenosine deaminase activity in human erythocytes by on-column Capillary isotachophoresis - capillary zone electrophoresis in the present of electrosmotic flow

  • Original language description

Transient capillary isotachophoresis - capillary electrophoresis at the presenseof electroosmotic flow was utilized for the measurement of adenosine deaminase activity in human erythocytes. Phosphates, a dominant anios of the sample matrice were used asleading ions for transint isotachophoresis and borates (0.3 M, pH 10) were used as terminating ions and background electrolyte for capillary zone electrophoresis. Final experimental conditions allowed to inject 70% of the total capillary volume (1.45 mL)with the sample. Product of enzymatic conversions present in the sample in low-micromoral range were determined by the use of optimized conditions. Limit of detection was 28 nM using UV detection at 202 nm. Presented data showed that CITP-CZE can be performed in uncoated capillaries at the presence of strong EOF.
