Original language description
The wild type human p53 protein binds preferentially to supercoiled DNA in vitro both in the presence and absence of p53 consensus sequence. The p53-DNA binding is decreased by physiological concentrations of zinc ions and at higher zinc concentrations the binding of p53 to sc DNA is completely inhibited. In contrast, oxidation of the protein with diamide resulted in partial inhibition of p53 - scDNA binding. The activation of p53 for sequence-specific binding within scDNA was also studied. C-terminal specific antibody Bp53-10 activated the sequence-specific binding of p53 to p53CON within pPGM1 scDNA but did not influence its binding to pBluescript scDNA.
