    Fluctuation of stem cell niche as a source of tissue adaptability

    At the most general level, stem cells are characterized as cells that are able to replicate (keeping their stemness) and, in parallel, their daughter cells are able to differentiate and build functional tissues. Although the description of stem cells dates back to the nineteenth century, their basic characteristic has remained in principle unchanged. In our work, we address the question to what extent stem cell behaviour is stable and predictable and to what extent it is fluid and erratic. Using continuously growing mouse teeth, model system for the study of stem cell dynamics, we investigate fundamental questions concerning the stem cell sources and activation and fluctuation of stem cell niche. Using state-of-the-art approaches including single-cell RNA-sequencing and lineage tracing, we bring new insights into stem cell behaviour on the fundamental level. Our work presents the stem cell niche as a highly adaptive microenvironment that is able to respond very efficiently to rapidly changing environmental conditions and thus to the actual organism's needs. These attributes of stem cells are accompanied by their less predictable behaviour and result in high plasticity at the cellular and tissue level. Taken together, our new theory of stem cell niche behaviour not only elucidates some mechanisms of continuous tissue regeneration at the cellular and molecular level, but also explains the high degree of adaptability to injury or other unpredictable tissue changes.
