Expression of putative mycobacterial glycosyltransferase Rv1526c

  • Original language description

At present, there can be seen a huge prevalence of tuberculosis all over the world. The most serious problem of its treatment arises specially from multidrugs resistant (MDR) forms of Mycobacterium tuberculosis patogen that are not possible to cure by conventional antimycobacterial therapies. Our research is focused on glycosyltransferases (GTs) from M. tuberculosis, enzymes that are responsible for biosynthesis of a cell wall envelope of this pathogen. They build some unique sugar components that playa crucial role in host-pathogen interactions. Using various bioinformatics tools applied to M. tuberculosis genome sequence, genes of some putative GTs were selected and cosmids carrying DNA of these GTs were obtained from Pasteur institute in Paris. Presented contribution is focused on putative rhamnosyltransferase Rv1526c. Generally, the most complicating point in preparation of GTs is low expression of these proteins in soluble form (proteins are toxic for expression host cells, proba
