Expression and purification of putative rhamnosyltransferase from Mycobacterium tuberculosis
Original language description
The resistance of Mycobacterium tuberculosis is related to an unusual and complex structure of mycobacterial cell wall envelope. The cell wall consists of a core composed of cross-linked peptidoglycan to which the heteropolysaccharide arabinogalactan isattached, the distal ends of which are almost esterified with mycolic acids (mAGP). The second of the major macromolecular compounds of mycobacterial cell wall is lipoarabinomannan (LAM) that has been considered as a key surface molecule in host-pathogeninteractions[1,2]. In contrary to the knowledge of the major cell wall constituents there is not much known about their biosynthesis pathways and glycosyltransferases (GT) involved. The global aim of the project is to understand the role of some key glycosyltransferases focusing on their structural analysis. Using various bioinformatics tools, some promising sequences in the M. tuberculosis genome were found. Presented work is focused on RfbE protein, a putative mycobacterial rhamnosylt
