Identification of European diplozoids (Diplozoidae, Monogenea) using the RFLP pattern of second internal transcribed spacer (ITS2)

Original language description

The second internal transcribed spacer (ITS2) of the ribosomal RNA genes of Diplozoon paradoxum and Paradiplozoon nagibinae were amplified and sequenced. The PCR product of D. paradoxum was bigger (840bp) than that of P. nagibinae (820bp). There was no intraspecific variability recorded in sequences from either species. Sequence comparisons and ITS2 RFLP pattern of eight European diplozoid species aimed to resolve their identification and amends the previous studies. Restriction fragment length polymorphism (RFLP) was used to distinguish both species from each other and from P. bliccae, P. homoion, P. megan, P. pavlovskii, P. sapae and Eudiplozoon nipponicum, using restriction enzymes AluI, HaeIII, HinfI, RsaI and SphI. ITS2 rDNA byla sekvenovana pro druhy Diplozoon paradoxum a Paradiplozoon nagibinae. PCR produkt D. paradoxum byl vetsi (840bp) nez pro P. nagibinae (820bp). Nebyla nalezena vnitrodruhova variabilita v sekvenci obou druhu. Porovnani sekvenci a vysledku RFLP ITS prokazalo existenci osmi druhu evropskych diplozoonu.
