Original language name
Variations in ITS of ribosomal DNA of Czech Armillaria species determined by PCR and high performance liquid chromatography
Original language description
We analysed 40 isolates of species A. borealis, A. cepistipes, A. gallica, A. mellea, A. ostoyae and A. tabescens mostly collected in Czech Republic by PCR-RFLP.Restriction fragments were analysed by ion-exchange high performance liquid chromatography that proved to be more useful, more sensible and less time consuming than classical electrophoresis on agarose gel.The cluster analysis based on the restrictions patterns of restriction endonucleases AluI and HinfI divided the analysed species into three groups.
Czech name
Determinace variability ITS regionu v rámci ribozomální DNA václavek pomocí PCR a HPLC
Czech description
Analyzovali jsme celkem 40 izolátů druhů A. borealis, A. cepistipes, A. gallica, A. mellea, A. ostoyae a A. tabescens pomocí PCr a následné restrikční anlýzy. Restrikční fragenty byly analyzovány pomocí HPLC kolony, což zvýšilo senzitivitu, přesnost, rychlost a snížilo cenu detekce.
