c-Myb is a key regulator of progenitor cells in bone marrow, colonic crypts and adult brain. Activation of c-myb is associated with leukemias and solid tumors. The aim of this study was to assess the role of the c-Myb protein in control of invasion of breast cancer cells. c-Myb overexpression enhanced migration and invasiveness of MDA-MB-231 cells as determined by transwell cell invasion assay and real-time analysis of cell migration and invasion using the xCELLigence RTCA system. qRT-PCR analysis revealed c-Myb-dependent upregulation of the genes coding for proteases that are involved in invasive behavior of MDA-MB-231 cells: increased production of cathepsin D and matrix metalloproteinase 9 (MMP9) in MDA-MB-231MYBup cells were further confirmed by western blotting. Transactivation assay revealed that c-Myb can activate transcription from the cathepsin D promoter.
