Original language description
Mycobacterium tuberculosis is a gram positive, acid resistent bacterium causing chronic illness. Tuberculosis is characterized by a disease persistence and overall difficulties associated with its treatment. A combination of the sophisticated defence barrier and the slow metabolism of the bacterium make it a difficult target for therapeutics. The mycobacterial cell wall is an essential, covalently linked complex of highly hydrophobic mycolic acids, peptidoglycan and heteropolymeric arabinogalactan. Arabinogalactan is composed of about 30 alternating 5- and 6-linked b-D-galactofuranose (Galf) units. It anchores in peptidoglycan by a dedicated linker unit, a-L-rhamnopyranosyl-(1-3)-N-acetyl-a-D-glucosaminyl-phosphate [Rha-(1-3)-GlcNAc-P]. Two to three arabinan chains containing approximately 23 D-arabinofuranose (Araf) units each, composed mostly of 5-linked a-D-Araf with branching introduced by 3,5-a-D-Araf, are bound to the galactan.
