Study of red clover (Trifolium pratense L.) genes specific for phytoalexin synthesis

  • Original language description

Phytoalexins are chemically diverse group of plant antimicrobial compounds, which are synthesized de novo by plants in response to biotic stress. Trifolirhizin is an isoflavonoid phytoalexin specific for red clover (Trifolium pratense L.). The aim of the study was experimental assessment of correlation between the expression of candidate genes and the level of trifolirhizin synthesized in the samples affected by biotic stress in comparison with control samples. Plant material included 4 red clover varieties (Tatra, Fresko, Tempus, Hungarotetra) and newly bred genotypes ( PO..) with different level or resistance to fungus Erysiphe trifolii. Three different types of stresses were applied.Computational analysis comprised in silico identification of four candidate genes for enzymes of biosynthetic pathway: phenylalanine ammonia lyase (PAL; EC4.3.1.5), chalcone synthase (CHS, EC 2.3.1.74), cinnamate 4-monooxygenase (CYP; EC 1.14.13.11) and 2'-hydroxyisoflavone reductase (IFR; EC 1.3.1.45). The analysis of gene PAL revealed increased expression in three plants influenced by pathogen infection and in one plant influenced by cutting off the leaves.
