AUXIN AND CYTOKININ CROSSTALK DURING DE NOVO ORGANOGENESIS IN ARABIDOPSIS THALIANA

  • Original language description

The long known phenomenon of hypocotyl regeneration suggests the principal role of two phytohormones, cytokinins and auxin in the plant de novo organogenesis. The identity of newly formed organs was shown to be dependent on different concentration ratiosof cytokinin and auxin. This work is focused on influence of auxin/cytokinin interaction on different markers using a hypocotyl experimental system. Several Arabidopsis lines with modulated cytokinin metabolism or signalling were used as an experimentalmaterial: Pro35S::AtCKX lines with depleted endogenous cytokinin levels and ahk single and double mutant lines (mutants in cytokinin receptors). In these lines auxin efflux proteins ? PINs, auxin influx proteins ? AUX/LAX, artificial auxin inducible promoter DR5, two component signalling sensor ? TCS, have been also used as markers. Pro35S::AtCKX lines, ahk single and double mutant lines were compared with the wild type during the first five days of cultivation.
