Application of immobilized trypsin reactors as proteomics tool. Techniques and procedures for disease diagnostics
Original language description
In proteomics study, trypsin is obviously the most often used immobilized proteinase for the preparation of peptide maps. We immobilized trypsin on cellulose nanofibers and on magnetic silica nanoparticles. Both enzyme reactors were applied for proteolytic digestion of model proteins (alfa-casein and bovine albumin). Analysis showed that immobilized trypsin is well usable and comfortable. There were good consensuses within protein recognition in comparison with soluble trypsin. Sequence coverage withinmagnetic silica nanoparticles functionalized by trypsin was higher than 47.6%. Sequence coverage for trypsin-nanofibers could not be determined due to high number of non-specific fragments.
