Use of HPLC and GPC in the characterization of biocompatible polymer degradation products
Original language description
New non-toxic biodegradable biocompatible amphiphilic end-functionalized hydrogel based on the thermosensitive triblock macromonomer gained from hydrophilic poly(ethylene glycol) and hydrophobic copolymer of poly(lactic acid-co-glycolic acid) modified byitaconic acid (PEG-ITA-PLGA-ITA-PEG) was synthesized and analyzed using liquid chromatography methods. GPC analysis was used for the determination of an average molecular weight and polymer distribution of the single steps of the biodegradable PEG-ITA-PLGA-ITA-PEG macromonomer synthesis. On the other hand, the RP-HPLC method was optimized for the determination of the degradation products of PEG-ITA-PLGA-ITA-PEG copolymer. The peaks separation was influenced by a mobile phase composition, pH, injectionvolume and a sample solvent. The best separation of the glycolic, diglycolic and lactic acid was obtained for the mobile phase consisted of binary solvent mixture of acetonitrile : phosphate buffer having ratio of 5:95 (v/v) at pH = 3 and
