Magnetic and non-magnetic solid polymer microsphere for DNA diagnostics of bacteria in complex samples
Original language description
Magnetically driven separation techniques using micro- and nanosized magnetic solid carriers have become common place in molecular diagnostics of microorganisms in recent years. The aim of this work was to study the potential inhibition effect of different types of commercial and non-commercial magnetic and non-magnetic carriers on PCR course. There were tested 25 types of nano-/micro-particles such as magnetic non-porous P(HEMA-co-GMA), P(HEMA-co-EDMA), PGMA particles covered by carboxyl groups, and thermosensitive non-magnetic poly(N-isopropylacrylamide) microspheres - P(NIPAAm)4 and etc. Real-time PCR (qPCR) was used for an estimation of the PCR interference by comparing the amplification efficiencies of purified DNA containing solid nanoparticles with the DNA standards free of any nanoparticles.
