In vitro amplification of nucleic acids using the polymerase chain reaction (PCR) has become a powerful diagnostic tool for the identification of pathogens or beneficial microorganisms (probiotics) in foods. The isolation of DNA from analysed samples isalways an integral part of a multistep identification procedure. High quality of isolated DNA (PCR-ready) is necessary for DNA amplification by PCR. Superparamagnetic particles have been attractive for molecular diagnostics applications due to their ability to interact with various biomolecules of interest. Manipulation with paramagnetic particles is easier than manipulation with non-magnetic ones. Reversible DNA adsorption on the surface of magnetic microparticles with carboxyl groups was used for theisolation of DNA. The lecture presents an overview of hydrophilic P(HEMA) based microspheres used as separation media for extraction and purification of DNAs in PCR-ready quality.
