Simple Liquid Chromatography system for Low Flow Rate Gradient Separations
Original language description
In this contribution we describe simple approach to the splitless micro-flow liquid chromatograph construction and we demonstrate the functionality of the proposed system. Separation system consists of the programmable syringe pump equipped with the strain gauge force transducer and the high pressure syringe connected to the selector valve via long capillary where mobile phase gradient is generated and stored. Operation of the system is controlled by LabVIEW development environment. Mobile phase gradient is created by successive sucking of defined volumes of four solutions with gradually changing composition into the capillary. Available linear gradients of volumes from 5 to 50 ?l traced by uracil are presented. Depending on the selected mode, the system is quickly pressurized and the syringe pump delivers the mobile phase at the constant flow rate or the LabVIEW application drives the syringe pump speed according to the signal from the force transducer and holds the pressure constant.
