A new apheresis procedure enables the collection of two WBC- reduced RBC units in a single donation. The aim of this study was to evaluate the quality parameters of apheresis WBC- reduced RBC units (A-RBC) including markers of hemolysis and red cells injury. We completed 20 double - erythrocytapheresis with the separator Haemonetics MCS+ and prepared 40 TU WBC ? reduced RBC. We investigated the content of Hb, Ht, WBC count and content of IgA in TU. Free Hb, K+, LDH, lactate, pH and annexin V were analyzed in the day 0 and 40. During the storage time free Hb increasing was higher in A-RBC, no differences in increasing K + value and no change in pH between A -RBC and WB ? RBC were founded. The LDH increasing was higher in A-RBC and correlated to hemolysis in both types of RBC, No correlation of K+ according to free Hb was found.The changes of annexin V were the same in both type of RBC, increase of annexin V levels correlated according to values of free Hb.

Nová aferetická technika umožňuje současný odběr dvou jednotek erytrocytů při jedné návštěvě dárce. Cílem práce bylo zhodnotit základní parametry jakosti erytrocytů z aferézy a markery hemolýzy a poškození buněk. Na separátoru Haemonetics MCS+ 20 erytrocytaferéz, při kterých jsme získali 40 transfuzních jednotek (TU) erytrocytů (EAD). Vyšetřili jsme: v den 0 hodnoty Hb, Ht, leukocytů a IgA v TU, v den 0 a 40 volný Hb, LDH, kalium, glukosa, laktát, pH a annexin V. Vzestup obsahu volného Hb a byl vyšší uEAD než u ERD, u obou typů přípravků byl srovnatelný vzestup kalia, a pokles pH. Vzestup hodnot LDH během skladování byl vyšší u EAD a koreloval se stupněm hemolýzy u obou typů erytrocytů. Vzestup laktátu během skladování byl vyšší u ERD. Korelaci kaliase stupněm hemolýzy jsme nepotvrdili. Vzestup annexinu V byl srovnatelný u obou typů TP, vyšší absolutní hodnoty u EAD korelovaly se vzestupem volného Hb.
