    Brachypodium distachyon (Bd) is a valuable model organism for studying interactions of cereals with pathogenic and non-pathogenic microorganisms. Host-pathogen interactions among Bd and plant pathogens have been previously described for several important cereal diseases. Our research focuses on the endophytic fungus Microdochium bolleyi (Mb) originating from wheat roots. Endophytic fungi do not cause visible symptoms on plants and have the potential in plant protection against pathogens. In general, research on endophytic fungi is complex, especially from a methodological point of view. There are several reasons why the endophytic fungus Mb and plant Bd creates an optimal model system for studying interactions. (1) The affinity of Mb and Bd is excellent, and the fungus grows readily in Bd tissues. (2) The inoculation method is simple, and (3) the structures of Mb are readily visible in host tissues by light microscopy. In addition, we have developed and optimized (4) specific primers for standard PCR and qPCR. (5) This fungus's dynamics of plant colonization are being thoroughly studied. In experiments, we can observe that (6) Bd plants inoculated with Mb showed significantly weaker symptoms of Fusarium culmorum infection on leaves than plants without endophyte. (7) Optimized methods and available primers from previous studies allow the study of gene expression of specific marker genes. The advantage of Mb over strictly biotrophic mycorrhizal organisms is (8) the possibility of cultivation on axenic media, which makes their use more suitable for eventual industrial production. Our studies show that B. distachyon can serve as a model host system for the endophytic fungus M. bolleyi.
