    3 University Symposium, 2025, 42. Brachypodium distachyon (Bd) serves as a valuable model organism for studying cereal interactions with both pathogenic and non-pathogenic fungi. We examined the endophytic fungus Microdochium bolleyi (Mb), originally isolated from wheat roots, and its colonization dynamics in Bd. Our findings confirm that Mb readily colonizes Bd tissues, with initial colonization occurring on germinating seeds and later spreading to the crown, stem base, and roots. The colonization process was visualized using light microscopy, and newly developed species-specific primers enabled precise detection and quantification of Mb. Notably, Bd plants inoculated with Mb exhibited significantly reduced symptoms of Fusarium culmorum infection compared to non-inoculated plants. Gene expression analysis revealed upregulation of key defense-related genes in plants after F. culmorum infection, though Mb-mediated resistance in Bd appeared to be independent of most tested genes. Given its ease of cultivation and compatibility with Bd, Mb represents an optimal model for investigating plant-fungal interactions and potential biocontrol strategies in cereals.
