Alfalfa (Medicago sativa L.) is a forage legume of world-wide importance used in agriculture. The genetic relationship and distance among cultivars is of great interest for breeding program. Utilization of diverse germplasm is needed to enhance the genetic diversity of cultivars. Morphological traits and Simple Sequence Repeat (SSR) were used to evaluate genetic variability of 14 alfalfa cultivars in Czech Republic. Six SSR primers generated 27 polymorphic bands. The number of alleles per locus ranged from 2 for B21E13 to 6 for MTIC451 and MTIC189, with an average 4.5 alleles. The average polymorphic information content (PIC) was 0.67, ranging from 0.37 to 0.8. The lowest and the highest PIC value were recorded for primer B21E13 and MTIC189, respectively. The genetic diversity based on morphological characters ranged from 7.1 to 31.1.
